摘要：A method of modifying a plant to increase vegetative growth of commerciallyvaluable plant structures at the expense of non-essential and non-commercial structuresis provided. A cDNA library of reproductive-bud mRNA of P. radiata is produced andscreened against cDNA prepared from vegetative buds, and non-hybridising colonieswere selected. Candidate clones were then selected for the presence of genes expressedin both male and female buds and which expressed on induction by gibberellin A4/7. Oneclone contained a cDNA corresponding to a gene having the characteristic of earlyappearance and highly specific expression in male and female reproductive bud
development, and the corresponding genomic DNA sequence including the tissue specificpromoter of the gene was obtained. The gene was modified by fusing a portion of thegene including the tissue-specific promoter of the gene with a structural gene for aribonuclease. Candidate gene constructs were used to transform Pinus radiata shootforming tissues. Explants from successful transformants were screened for expression ofthe ribonuclease gene upon induction.
